Biocompatibility and radioprotection by newly characterized melanin pigment and its production from Dietzia schimae NM3 in optimized whey medium by response surface methodology

Abstract Purpose To characterize and optimize the productivity of melanin using an extremotolerant actinobacterium, Dietzia schimae NM3, for first time. Methods An extracellular brown pigment produced by D. NM3 in nutrient broth cheese whey medium adding L-tyrosine. The extracted was analyzed UV-visible, HPLC, FTIR assays. radical scavenging activity (by DPPH) sun protection factor (SPF) were measured. cytotoxicity assayed MTT chromate biosorption measured atomic absorption spectroscopy. Finally, production optimized response surface methodology (RSM) Box-Behnken design medium. Result purified showed similar peak to standard (SIGMA) at 3.5 min C=O bands, NH 2 , CH, C-N, aromatic groups FTIR. SPF obtained 188.9% 20.22, respectively. Using assay, revealed non-toxic effect on normal human fibroblast (HFB) cell culture. yield 790 mg l ?1 tyrosinase 3400 U ml contained powder [5% (w v )], L-tyrosine 2.5 g CuSO 4 0.013 pH 10.5, incubated 32 °C 3 days. ANOVA results indicated significant P -value, model F probability, with insignificant lack fit. After optimization mono-factors, came up as 1.2 4040 . Conclusion This is report this natural valuable biological properties such high antioxidant radioprotection biocompatibility line.
